What is Hydrea used for?

Hydrea capsules 500mg are used to treat various cancers including melanoma, resistant chronic myelocytic leukaemia (CML), and recurrent carcinoma of the ovary that has become metastatic (spread to tissues away from the original tumour) or is inoperable.  Hydrea capsules 500mg are also used in addition to irradiation therapy to control primary local tumours of the skin in the head and neck area (excluding the lip), known as squamous cell (epidermoid) carcinomas; also to control carcinoma of the cervix in addition to irradiation therapy.  Depending on the type of tumour, Hydrea capsules 500mg are used either intermittently or continuously and work by the same mechanism of action to kill the tumour cells.

How does Hydrea work?

Hydrea capsules 500mg contain hydroxycarbamide, a cytotoxic drug used for chemotherapy to treat certain cancers.  Hydroxycarbamide in Hydrea capsules 500mg is an antimetabolite, which means it is incorporated into DNA during cell division, which causes immediate inhibition of DNA synthesis by blocking the action of the enzyme ribonucleotide reductase; but has no effect or RNA or protein synthesis.  This action of hydroxycarbamide in Hydrea capsules 500mg prevents the normal process of cell replication and repair in tumour cells, particularly cells that are normally resistant to irradiation therapy or have been damaged but not killed by irradiation therapy.  When used in combination with irradiation therapy Hydroxycarbamide in Hydrea capsules 500mg is thought to arrest the cell cycle of the tumour cells in the G1 or pre-DNA synthesis stage, so that they are more likely to be killed by irradiation therapy.  Hydrea capsules 500mg cause cell death in various cancers including both solid tumours cancers of the blood. 

What does Hydrea contain?

Hydrea capsules 500mg contain the active ingredient hydroxycarbamide or hydroxyurea that is used for chemotherapy to treat various solid tumours and cancers of the blood.  They also contain citric acid, gelatine, lactose, magnesium stearate, sodium phosphate, titanium dioxide and, capsule colorants.

What are the side effects of Hydrea?

The most commonly reported side effects when taking Hydrea capsules 500mg include bone marrow failure, causing reduced number of white blood cells (CD4 lymphocytes and neutrophils), decreased platelet count (thrombocytopenia) and anaemia; gastrointestinal symptoms including anorexia, stomatitis (inflammation in the mouth), nausea, vomiting, diarrhoea, and constipation; skin disorders including rash, redness, ulceration, hyperpigmentation, atrophy and nail disorders; fever, chills, malaise, male fertility problems (low or no sperm count), kidney problems due to increased blood uric acid levels, drowsiness.

When should Hydrea not be used?

You should not use Hydrea capsules 500mg if you:

  • are allergic to hydroxycarbamide or any ingredients in Hydrea
  • are pregnant or are breastfeeding
  • have kidney disease
  • have depressed bone marrow function with leukopenia or thrombocytopenia or severe anaemia
  • are receiving vaccination with a live virus as this may result in a severe infection
  • are taking medicines that interact with Hydrea including other myelosuppressive agents, other chemotherapy drugs like cytarabine, gout medications like probenecid, antiretroviral  medications for HIV like didanosine and stavudine, live vaccines

What medications interact with Hydrea?

Several medications interact with Hydrea capsules 500mg and should either not be taken while you are taking Hydrea or only after discussion and instruction from your doctor:

  • Drugs that cause a serious reaction with Hydrea: other myelosuppressive agents, other chemotherapy drugs like cytarabine, antiretroviral  medications for HIV like didanosine and stavudine, live vaccines
  • Drugs that are affected by Hydrea: other chemotherapy drugs like cytarabine,

How should Hydrea be taken?

You should take your Hydrea capsules 500mg swallowed whole with a glass of water, with or without food, according to the type of tumour being treated and the treatment regime recommended by your doctor.  If you have difficulty swallowing a capsule, your doctor may suggest that you empty the contents of the capsule into a glass of water and swallow immediately.  For continual therapy you should take your Hydrea capsules 500mg once daily.  If you are also having irradiation therapy you should take your Hydrea capsules 500mg at least 7 days prior to starting irradiation therapy and then every 3 days subsequently.  For intermittent therapy you should take your Hydrea capsules 500mg every 3 days.  You should continue to take your Hydrea capsules 500mg for as long as recommended by your doctor, which will depend on how well you respond and the side effects.

Missed dose of Hydrea

If you miss a dose of Hydrea capsules 500mg take it as soon as you remember, unless it is time to take the next dose, then skip the missed dose.  Do not take a double dose.

How should Hydrea be stored?

You should store your Hydrea capsules 500mg below 25°C in a cool dry place, avoid heat and keep the bottle tightly sealed.
